摘要 根据Jean-Dupouy-Camer所报道1.7kb基因序列而设计合成的引物,对中国九个地区的旋毛虫株的肌组织旋毛虫DNA进行了聚合酶链反应(PCR)。扩增产物经琼脂糖凝胶电泳分析,可见中国猪源旋毛虫均扩增出特异性的602bp和230bp大小的DNA条带,而中国犬源和猫源以及正常对照肌组织DNA均未扩增出特异性片段。本法对中国猪源旋毛虫可检测到0.02条旋毛虫DNA,具有高度的特异性和敏感性。 A pair of primers were designed and synthesized based on Jean Dupouy-Camet 1.7-kb DNA sequence and used to detect larvae of T.isolates. They were isolated from pigs of Harbin Hailun, Shenyang,Henan Nanyang,Hubei Shiyan,XiAn,Yunnan Dali and dogs of Harbin Wuchang,Changchun and cats of Heilongjiang Sunwu respectively. The amplified products were analysed by agarose gel electrophoresis and the specific 602 and 230 bp bands shown in the isolates of larvae T.spiralis from pig, while no band appeared in isolates from dog and cat and normol mice.The parasite density detected by this method could be as low as 1 parasite, suggesting that PCR techniqe has high sensitivity and specificity.
