摘要 应用高效液相色谱法分离测定玉米类胡萝卜素。结果表明:采用体积比为15:75:10的己烷、丙酮、水在室温暗处浸提48h的制备样品方法,使用Nova-pakC18不锈钢柱,以体积比为83:15:2的乙腈、三氯甲烷、叔丁醇作流动相,流量线性梯度洗脱,柱温25℃等色谱条件,在440nm波长下检测,使玉米中的主要类胡萝卜素在10min内得到了较好的分离。并以外标定量法对玉米中的4种主要类胡萝卜素进行了定量分析。 The carotenoids from corn grain were separated and determined by HPLC. Results showed that the main carotenoids in corn grain can be successfully separated in 10 min by employing Nova-pak C 18 stainless steel column, using acetonitrile-chloroform-tertiary butyl alcohol (volume ratio 83∶15∶2) as flow phase, applying linear gradient eluting, and using the column temperture of 25 ℃ and the detection wavelength of 440 nm after extracting the sample in hexane-acetone-water(volume ratio 15∶75∶10) for 48 h at room temperature in dark. Four kinds of main carotenoids from corn grain were determined by external standards.
