摘要
目的探讨竹叶总黄酮对心肌缺血再灌注损伤的保护作用及机制。方法60只SD大鼠随机分为三组,竹叶总黄酮组腹腔注射竹叶总黄酮,假手术组和缺血再灌注组腹腔注射等量生理盐水,连续7d,末次于冠脉结扎前2h腹腔注射给药。结扎部位位于左冠状动脉前降支中上1/3处,结扎1h后剪开结扎线,用止血钳夹闭胸腔,再灌注2h,检测心肌组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,检测血清LDH和CK水平。TTC染色测定心肌梗死面积。结果与缺血再灌注组比较,竹叶总黄酮MDA、LDH和CK含量显著下降,SOD活性明显升高,心肌梗死面积显著减少。结论竹叶总黄酮对大鼠心肌缺血再灌注损伤具有保护作用,其机制可能与增加SOD活性、增强心肌抗氧化能力、稳定心肌细胞膜电位有关。
Objective: To investigate the protective effects and its mechanism of bamboo leaves flavonoid on myocardial ischemia reperfusion injury (MIRI) in rats. Methods: 60 SD rats were divided randomly into 3 groups. The bamboo leaves flavonoid group was given bamboo leaves flavonoid in peritoneal. The Sham - operated group and the ischemia - reperfusion group were taken equivalent saline in peritoneal. After 7 days, these rats were injected drugs at the last time 2h before their coronary artery ligated. The ligation location is at the middle and superior 1/3 part of anterior descending branch of rat's left coronary artery. Ligatures were opened lh later. These rats'chests were closed by hemostat immediately, then they were given reperfusion 2h. The SOD activities, the MDA contents of the cardiac, the level of LDH and the level of CK were investigated. Meanwhile, the myocardial infarct size was detected by TTC staining. Results: Compared with the ischemia - reperfusion group, the SOD activity of bamboo leaves flavonoid group increased significantly. The MDA, the LDH and the CK contents of the cardiac muscle and the infarct size in bamboo leaves flavonoid group were decreased obviously. Conclusion: bamboo leaves flavonoid has the protective effect on myocardial ischemiareperfusion injury in rats. The mechanism of this effect could relate to the increasing SOD activity, enhancing the ability of antioxidation and stabilizing the cell transmembrane potential of cardiac muscle.
