芪天滴丸主要活性成分的大孔吸附树脂分离纯化工艺研究 被引量:6

Separation and Purification of Main Active Composition of Compound Qitian Dropping Pill by Macroporous Resin

摘要 目的采用大孔吸附树脂法,优化芪天滴丸中赤芍、红花及红景天活性成分的纯化工艺条件。方法以芍药苷、羟基红花黄色素A(HSYA)及红景天苷为指标,对4种树脂进行了筛选;以芍药苷和HSYA为指标,优化HPD100型大孔树脂富集纯化的工艺条件。结果HPD100型对芪天滴丸中芍药苷、HSYA及红景天苷具有很好的吸附特性,优化HPD100型大孔树脂富集纯化的工艺条件为:样品溶液含芍药苷2.98 g.L-1,含HSYA 1.44 g.L-1,含红景天苷0.81 g.L-1,吸附时间不低于12 h,吸附速度60 mL.h-1,洗脱速度60 mL.h-1,20%乙醇洗脱200 mL。大孔吸附树脂反复使用7次。结论HPD100大孔吸附树脂能有效富集纯化赤芍、红花及红景天中的活性成分。 OBJECTIVE To study the purification of the active composition from radix paeoniae rubra, safflower and rhodiola in Qitian Dropping Pill with macroporous resin. METHODS Firstly the kind of resin was screened by using hydroxysafflor yellow A, paeoniflorin and salidroside as index; then, the technical conditions of the enrichment and purification of HPD100 resin were optimized. RESULTS The HPD100 was used for adsorbing hydroxysafflor yellow A and paeoniflorin. The optimal conditions were obtained that the concentrations of sample extract of hydroxysafflor yellow A, paeoniflorin and salidroside was 2.98 g·L^-1, 1.44 g·L^-1, and 2.98 g·L^-1 respectively. Absorption flow rate was 60 mL·h^-1. Absorption time was above 12h, 20% alcohol was as the elution with rate of 60 mL·h^-1 and volume was 200 mL. The absorption ability resin was not decreased after being used repeatedly seven times. CONCLUSION The HPD100 is effective for the enrichment and purification of the active composition of radix paeoniae rubra, safflower and rhodiola.
