N-乙酰半胱氨酸对糖尿病肾病大鼠炎症反应的抑制作用 被引量:2

摘要 目的:探讨N-乙酰半胱氨酸(NAC)对糖尿病肾病(DN)大鼠炎症反应的抑制作用。方法:制备DN大鼠模型,将动物随机分为正常对照组、DN组和NAC组。8周后测定尿白蛋白排泄率,血清高敏C反应蛋白(hs-CRP)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)水平,免疫组化方法检测肾组织NF-κB蛋白表达水平,观察肾脏病理标本。结果:①与正常对照组比较,DN组和NAC组尿白蛋白排泄率增多(P<0.01),血清hs-CRP、IL-6和TNF-α水平升高(P<0.01或P<0.05),肾组织NF-κB蛋白表达水平升高(P<0.01或P<0.05),肾脏病理改变明显;②与DN组比较,NAC组尿白蛋白排泄率减少(P<0.01),血清hs-CRP、IL-6和TNF-α水平降低(P<0.05),肾组织NF-κB蛋白表达水平降低(P<0.05)和肾脏病理改变较轻。结论:NAC对DN大鼠肾脏有保护作用,其部分机制与抑制炎症反应有关。 Objective:To investigate the inhibition of N-acetylcysteine (NAC) on the inflammation of diabetic nephropathy rats. Methods:A rat model of DN was established. The rats were divided into control group, DN group and NAC group randomly. Urinary albumin excretion rate (UAER),hs-CRP,IL-6 and TNF-α in serum were determined. The expression of NF-κB in renal tissue was detected by immunohistochemistry 8 weeks after treatment. Pathomorphologic changes of nephridial tissues were observed. Results:① Compared to control group, UAER, hs-CRP, IL-6 and TNF-α in serum were significantly higher(P 〈 0.01 or P 〈 0.05), activity of NF-κB in nephridial tissue increased (P 〈 0.01 or P 〈 0.05) and significant changes of pathomorphology in nephridial tissues were observed in DN group and NAC group. ②Compared to DN group, UAER,hs-CRP, IL-6 and TNF-α in serum were significantly lower(P 〈 0.05), activity of NF-κB in nephridial tissue was lower (P 〈 0.05) and improvement of pathomorphology in nephridial tissues were observed in NAC group. Conclusion:The protective effect of NAC on the kidney of the DN rats may be partly related with the inhibition on inflammation.
