摘要
目的探讨原白头翁素衍生物(溴甲基呋喃酮)体外对乳腺癌MCF-7细胞株的抗增殖及诱导凋亡作用。方法应用MTT法、倒置显微镜、HE染色、扫描电镜技术、吖啶橙/溴化乙锭荧光染色方法以及流式细胞技术检测原白头翁素衍生物对体外培养的人乳腺癌MCF-7细胞的诱导凋亡及细胞形态学改变。结果MTT法示不同浓度原白头翁素衍生物对MCF-7细胞株均有抗增殖作用,并呈时间—浓度依赖性。倒置显微镜、HE染色、扫描电镜观察示核固缩、碎裂、凋亡小体形成等典型形态学改变。AO/EB双染色结果显示细胞死亡主要以凋亡为主。流式细胞示随着药物浓度的增加细胞凋亡率亦逐渐升高,S期细胞所占比例增大。结论原白头翁素衍生物在体外对人MCF-7乳腺癌细胞株有抗增殖及诱导凋亡作用。
Objective To investigate the effects of isomycin derivant on anti-proliferation and inducing apoptosis of human breast carcinoma cell line. Methods MTF assay was used to measure the growth inhibition rate of human breast carcinoma MCF-7 cell line treated with isomycin derivant. Morphological changes of the apoptosis were observed by using inverted microscope and HE staining. AO/EB double staining was applied to discriminate the apoptotic cells from dead ones. Scanning electron microscope was used to observe the ultrastructure changes. Flow cytometry was used to investigate the apoptosis and changes of cell cycle. Results MTY assay showed that isomycin derivant could inhibit the proliferation in a dose- and time-dependent manner. Morphological changes included cell nucleus condensation, plasma bleb formation and the apoptotic body formation. Flow cytometry showed an inducing apoptosis and interfering cell cycle results. Conclusion Isomycin derivant is useful for the anti-proliferation and apoptosis of breast carcinoma cells.
