杉木茎尖诱导愈伤组织及植株再生研究 被引量:21

摘要 用杉木优良单株基部萌蘖条茎尖作为外植体进行组培快繁试验,结果表明:愈伤组织在改良MS+KT 0.2 mg/L+2,4-D 1.0 mg/L培养基上,外植体诱导率为86.7%;不定芽分化与增殖培养基为改良MS+KT 1.0 mg/L+IBA 0.5 mg/L,分化系数达3.42;芽苗培育约30 d后,用1/2 MS+IBA 0.2 mg/L+NAA 0.5 mg/L作生根培养基培养约25 d,生根率可达93%以上。生根苗移植到覆盖3 cm厚黄心土的苗床上栽培,成活率可达90%以上。 The shoot tips sprouted from the trunk base of the superior individual plants of Cunninghamia lanceolata were applied as the explants to induce the callus on the improved medium MS + KT 0. 2 mg/L + 2,4 - D 1.0 mg/L. The results showed that the induction rate reached 86.7% ; the culture medium for differentiation and multiplication of adventitious bud was the improved MS + KT 1.0 mg/L + IBA 0.5mg/L,whose differentiation coef- ficient reached 3.42. The culture medium for rooting to be applied 30 days later was the improved 1/2MS + IBA 0. 2 mg/L + NAA 0. 5 mg/L, by which the rooting rate was 93% within 25 days. The rooted seedlings were trans- planted to the seedbed covered by 3 cm yellow soil for further cultivation, and the transplanting survival rate was over 90%.
