摘要
目的:筛选可用于胃癌的诊断及手术后疗效评估的特征性蛋白质分子标志物。方法:采用表面增强激光解吸离子化-飞行时间质谱(SELDI-TOF-MS)技术,对胃癌、胃癌术后、慢性胃炎患者和健康对照组蛋白质指纹图谱的分析,同时采用多肿瘤标志物蛋白芯片检测系统测定各组血清CA19-9、CEA、CA125的表达情况。结果:胃癌与正常人血清蛋白指纹图谱,共有6种蛋白质表达量有明显差异。利用SELDI-TOF-MS技术组建的筛选模型检测胃癌敏感度为89.9%,特异性为93.1%,阳性和阴性结果预测值均为100%,对胃癌的诊断率为92%。结论:利用SELDI-TOF-MS技术在血清中可以筛选到诊断胃癌的特异性蛋白质标志物,并可对胃癌术后、预后情况以及肿瘤的再次复发提供实验室诊断依据,该技术的诊断性能优于常规肿瘤标志物,并为胃癌的早期治疗提供了可能。
Objective: To screen for gastric cancer diagnosis and assessment of the characteristics of post-operative effect of protein molecular markers. Methods: The surface-enhanced laser desorption ionization - time of flight - mass spectrometry (SELDI-TOF-MS) technology, gastric cancer, gastric cancer, chronic gastritis patients and healthy controls of protein fingerprint analysis, while a multi-tumor markers protein chip detection system in each group of serum CA19-9, CEA, CA125 expressien. Results: The gastric cancer and normal serum protein fingerprinting, a total of six kinds of significant differences in the amount of protein expression using SELDI-TOF-MS technology to establish a screening model for testing the sensitivity of gastric cancer 89.9%, specificity of 93.1%, positive and negative results predictive value were 100% in gastric cancer diagnosis rate was 92%. Conclusion: The use of SELDI-TOF-MS technology in the serum can be screened to the diagnosis of gastric cancer-specific protein markers, it may also gastric cancer, prognosis and tumor recurrence again to provide laboratory diagnosis based on diagnostic performance of the technology priorities conventional tumor markers, and to provide early treatment of gastric cancer possible.
