摘要 目的:检测Versican核心蛋白及其mRNA在胎鼠磨牙牙胚发育早期的表达,探讨其在牙胚发育过程中的作用。方法:制备胎鼠磨牙牙胚各发育阶段标本,用免疫组化和原位杂交方法分析Versican核心蛋白及其mRNA在牙胚组织中的表达与分布。结果:Versican核心蛋白及其mRNA在胎鼠磨牙牙胚不同发育阶段表达各异,在细胞增生活跃区如增生的牙板、内外釉上皮细胞、颈环以及牙乳头等区域表达明显,而在已分化细胞如成牙本质细胞、成釉细胞等区域无表达,在星网状细胞所在区域也未检测到。结论:Versican核心蛋白及其mRNA在牙胚发生过程中呈时间-空间特异性模式表达。它可能与上皮-间充质相互作用以及相关细胞的增殖和分化有关。 Objective:To study the expression of Versican core protein and its mRNA in the embryonic molar tooth germ of mice.Methods:5 μm serial paraffin sections of the first mandibular molars at different development stages were cut.The expression pattern of Versican core protein and its mRNA were analyzed by immunohistochemistry and in situ hybridization respectively.Results:The expression and localization of Versican core protein and its mRNA varied at different stages,and intensely expressed in the proliferation cells such as outer and inner enamel epithelium,dental lamina,dental papillae as well as cervical loops.There was no expression at the differentiated cells including the odontoblasts and ameloblasts,as well as the stellate reticulum cells.Conclusion:The expression of Versican is in a temporal-spatial specific pattern.It may be involved in the epithelial-mesenchymal interaction,particularly in cell proliferation and differentiation at the embryonic molar tooth germs.
