一种季铵盐型抗菌单体对小鼠成纤维细胞的体外细胞毒性 被引量:3

摘要 目的:评价一种抗菌高分子单体对小鼠成纤维细胞的细胞毒性。方法:配制浓度分别为100、60、40、20、10、5、2、1、0.5μg/ml的抗菌单体(甲基丙烯酰氧乙基-正十六烷基-二甲基氯化铵,DMAE-CB)溶液,采用甲基噻唑基四唑比色法(MTT法)对小鼠成纤维细胞的细胞毒性进行比较。结果:抗菌单体浓度在100、60、40、20、10、5、2μg/ml时,细胞的相对增殖率与空白对照组之间均存在显著性差异(P<0.05)。依据相对增殖率计算得到抗菌单体的LC50为3.02μg/ml,与常用基质单体双酚A双甲基丙烯酸缩水甘油酯(Bis-GMA)的细胞毒性相当。结论:尽管作为单体形式存在时,DMAE-CB具有较大的细胞毒性,但仍具有将其键合于树脂基材料中发挥接触抗菌作用的良好前景。 Objective:To evaluate the in vitro cytotoxicity of a quaternary ammonium salt antibacterial monomer(methacryloxylethyl cetyl ammonium chloride,DMAE-CB) on mouse fibroblasts.Methods:The solutions of quaternary ammonium salt monomer of 100,60,40,20,10,5,2,1,0.5 μg/ml were prepared.The in vitro cytotoxicity was measured by MTT method.Results:No cytotoxic effect was observed on contact with DMAE-CB at concentrations of 1 μg/ml or less.The median lethal concentration(LC50) for L929 which was calculated according to relative growth rate of cells was 3.02 μg/ml.The cytotoxicity of DMAE-CB was considered to be similar with that of dental matrix monomer Bis-GMA.Conclusion:The results suggest that the cytotoxicity of DMAE-CB is comparable with that of dental resin matrix.Therefore,incorporating DMAE-CB with dental resin-based materials does not increase the cytotoxicity.
