基质金属蛋白酶-8,11,19和组织蛋白酶B在异常胎盘绒毛组织中的表达 被引量:3

Expression of matrix metalloproteinase-8,11,19 and cathepsin B in abnormal placental tissues

摘要 目的:探讨基质金属蛋白酶-8,11,19和组织蛋白酶在胎盘绒毛组织中的表达差异与葡萄胎、子痫前期等和滋养层细胞密切相关的妊娠疾病病理机制的关系。方法:采用RT-PCR检测方法,比较葡萄胎组织、早孕绒毛、子痫前期足月胎盘和自然分娩及剖宫产的正常足月胎盘组织中基质金属蛋白酶-8,11,19和组织蛋白酶B等蛋白酶的表达差异。结果:5种胎盘绒毛组织中,仅早孕绒毛明显表达基质金属蛋白酶-8(P<0.05),而基质金属蛋白酶-11的表达在足月胎盘组织中较为明显(P<0.05),且子痫前期胎盘组织中基质金属蛋白酶-11的表达较正常胎盘组织弱(P<0.05)。葡萄胎组织中基质金属蛋白酶-19的表达弱于正常早孕绒毛组织(P<0.05),而其在子痫前期胎盘组织中的表达强于正常自然分娩的胎盘组织(P<0.05)。5种绒毛组织中组织蛋白酶B的表达比较差异无统计学意义(P>0.05)。结论:基质金属蛋白酶-8,11,19等基质金属蛋白酶在胎盘绒毛组织中的表达变化,可能和葡萄胎、子痫前期等与滋养层密切相关的妊娠疾病的病理发生有关。 Objective To study the relationship between the expression of invasion-associated proteinases and the pathogenesis of hydatidiform mole or preeclampsia which is intensively correlated with trophoblast.Methods The semi-quantitative method of RT-PCR was employed to detect the expressions of matrix metalloproteinases-8,-11,-19 and cathepsin B in tissues of hydatidiform mole,the first trimester villi,normal term placenta(parturition or cesarean)and placenta with preeclampsia.Results Among the 5 kinds of tissues,the expression of matrix metalloproteinase-8 was only seen in the first trimester villi.The expression of matrix metalloproteinase-11 was significant in normal and abnormal term placenta(P0.05),while the mRNA level of matrix metalloproteinase-11 was lower in tissues with preeclampsia compared to normal term placenta(P0.05).The expression of matrix metalloproteinase-19 mRNA in hydatidiform mole tissues was lower than in the first trimester villi(P0.05).However,it was higher in placenta with preeclampsia than in normal term placenta.No difference was found in the expression level of cathepsin B in these tissues between each other(P0.05).Conclusion The changes of matrix metalloproteinase-8,-11 and-19 expressions in placenta tissues might be involved in the pathogenesis of trophoblast-associated diseases of pregnancy such as hydatidiform mole and preeclampsia.
