实时荧光PCR方法对乙肝病毒基因分型的检测及临床意义

Detection of hepatitis B virus gene type with RT-PCR method and its clinical significance

[目的]了解乙肝患者的HBV基因分型特点及不同乙型肝炎病毒基因型对肝脏的损害程度,了解不同乙肝基因型的临床用药效果。[方法]随机选取乙型肝炎患者血清100例,已确诊的慢性乙肝血清50例,肝硬化患者的血清20例和追踪治疗的乙肝患者血清15例。用实时荧光PCR(RT-PCR)方法对其进行基因分型的检测。[结果]在100例乙肝患者中,B型20例,占20%;C型71例,占71%;BC混合型1例,占1%;非B非C型8例,占8%。肝硬化患者中,以C型为主。[结论]乙肝基因型主要以C型为主,B基因型与C基因型的临床治疗效果比其他基因型差,且C基因型感染的患者肝硬化发生率高。

[Objective] To understand the character of hepatitis B virus genotype,know the damage of liver in different hepatitis B virus genotype infection,and observe the curative effects of medications on different hepatitis B virus genotype infection.[Methods] Random choose serums from 100 patients with HBV infection,50 patients diagnosed as chronic hepatitis B,20 patients with liver cirrhosis and 15 hepatitis B patients that have been treated.Their genotype was detected with real-time PCR method.[Results] In 100 samples with HBV infection,20% of them were genotype B,71% were genotype C,1% was mixed type,and 8% of the samples were non-B and non-C.Genotype C was predominant genotype in liver cirrhosis.[Conclusion] Genotype C is predominant genotype.The treatment result in genotype B and genotype C is worse than that in other types.The incidence rate of liver cirrhosis is high in genotype C infection.
