MAE-分光光度法测定大白口蘑中总黄酮含量 被引量:1

摘要 [目的]采用微波辅助萃取(MAE)法提取、以分光光度法测定大白口蘑样品中的总黄酮含量。[方法]通过单因素试验和正交试验考察乙醇浓度、液固比、微波功率、微波时间等因素对微波辅助萃取法提取大白口蘑黄酮提取率的影响。对微波法与乙醇浸提法、超声波萃取法进行了比较。[结果]微波萃取法最佳提取条件:乙醇浓度60%、液固比40∶1、微波功率255 W、微波时间5 min。最优条件下,精密度和回收率试验所得结果的相对标准偏差为1.24%(n=6),回收率在98.1%~103.7%。微波法优于乙醇浸提法和超声波萃取法。[结论]MAE-分光光度法简单、可靠,重现性好,稳定性好,可广泛用于各种食用菌中黄酮类物质含量的测定。 [ Objective ] The research aimed to determine the content of total flavones from Tricholoma giganteum by microwave-assisted extraction and spectrophotometry. [ Method] The effects of solvent concentration, ratio of solution to solid, microwave power and radiation time on the extraction rate of flavone from T. giganteum by microwave-assisted extraction were studied by single factor and orthogonal experiment. The microwave method, ethanol extraction method and ultrasonic extraction method were compared. [ Result ] The extraction conditions of flavones from Tricholoma giganteum by microwave-assisted method were optimized as 40:1 of the liquid-solid ratio,60% ethanol to twigs and microwave power of 255 W for 5 min. Under the optimized conditions ,the relative standard deviation was 1.24% ( n = 6 ) and the recovery was 98.1% - 103.7%. Microwave method was better than ethanol method and ultrasonic extraction method. [ Conclusion] MAE-spectrophotometry was simple and accurate with good reproducibility and stability. It can be used for the determination of flavones in many kinds of edible fungi.
