丁苯肽软胶囊对大鼠局灶性脑缺血后梗死体积和神经元凋亡相关蛋白bcl-2,caspass-3表达的影响 被引量:8

摘要 目的观察丁苯肽软胶囊(商品名:恩必普,NBP)对大鼠局灶性脑缺血后梗死体积和神经调亡相关蛋白bcl-2;Caspase-3表达的影响。方法将SD大鼠随机分为缺血对照组、NBP大、中、小剂量组,采用大鼠大脑中动脉栓塞模型以及氯化三苯四唑(TTC)染色和免疫组织化学染色方法,在不同时间点观察各组大鼠脑梗死体积的变化,以及Bcl-2、Caspase-3阳性细胞表达的不同。结果在不同时间点各给药组大鼠的脑梗死体积小于缺血对照组,有统计学意义(P<0.05);各给药组大鼠bcl-2阳性细胞表达的数目均高于缺血对照组,而caspase-3阳性细胞表达数目则低于缺血对照组(P<0.05);NBP大、中剂量组有一定的量效关系。结论NBP可减少脑梗死体积,在脑缺血损伤急性期可增加bcl-2的表达而减少Caspase-3的表达。 Objective To investigate the effects of Dingbentai s6ft capsule on bcl-2 and easpase-3 expression of apoptosis of neurons. Methods SD rats were randomly divided into ischemia control group, large-dose group, middle-dose group and low-dose group. The middle cerebal artery of rats was occluded for 2h by inserting an intraluminal filament,and reperfusion was then performed for 4h or 22h. The brains were stained with TTC for assessment of volume of infarction, and then embedded onto slides with paraffin for morphological assessment and immunohistochemistry was carried out to investi- gate the changes in bcl-2 and caspase-3. Results All treated groups at different times decreased the volume of infarction( P 〈 0.05). All treated groups at different times increased bel-2 and decreased cpspase-3 expression as well,while large-dose group showed more distinct effect( P 〈 0.05 ). Large-dose group and middle-dose group had positive dose-effect relation. Conclusion Dingbentai soft capsule can reduce the volume of cerebral infarction, and increase bcl-2 and decrease caspase- 3 expression.
