HHcy大鼠学习记忆能力和基底前脑ChAT的变化及早期叶酸干预 被引量:1

摘要 目的观察高同型半胱氨酸血症(HHcy)对大鼠学习记忆能力和基底前脑胆碱乙酰基转移酶(ChAT)的影响及早期叶酸(FA)干预的作用。方法将Wistar大鼠随机分为对照组、HHcy组和FA组,对照组给予饮用水,在HHcy组饮用水中添加蛋氨酸1.0g/(kg·d),在FA组饮用水中添加蛋氨酸1.0g/(kg·d)并予FA0.4mg/(kg·d)灌胃,共进行8周,测定各组血浆同型半胱氨酸(Hcy)、FA的基础浓度及实验满8周时的浓度,Morris水迷宫实验评估各组的学习记忆能力,并对大鼠基底前脑进行ChAT的免疫组化实验,对其灰度值进行分析。结果HHcy组的学习记忆能力较对照组和FA组均减退(P<0.01),经灰度分析比较后显示HHcy组基底前脑隔内侧核、Meynert’s基底核ChAT阳性细胞的表达与其余两组比较差异有显著性(P<0.01)。结论过量的蛋氨酸饮食可致HHcy,影响学习记忆能力,HHcy影响学习记忆能力考虑与基底前脑核团的胆碱能神经元的减少有关;早期补充叶酸可降低血浆Hcy水平,改善HHcy对学习记忆能力的影响。
