摘要
研究颈内动脉灌注基因重组葡激酶对脑血栓的溶化作用及对出、凝血和血液纤溶系统的影响。方法制成猪血栓性脑栓塞模型,4h后自颈内动脉灌注不同剂量的重组葡激酶(0.1、0.2、0.4mg/kg),脑血管造影观察血栓是否溶化,并测定出、凝血及纤溶系统指标。结果随葡激酶剂量增大,血管再通率增高,大剂量组对出、凝血及纤溶系统的影响较明显。结论重组葡激酶能有效溶化猪脑血栓,最佳剂量为0.2mg/kg。
PURPOSE To assess the thrombolytic efficacy and safety of recombinant staphynokinase(rSaK) by intra arterial infusion in a experimental embolic stroke model in pig. METHODS We established a pig model of thromboembolic stroke by injecting fragments of human plasma clot into one internal carotidartery. Four hours after embolization, the pigs were blindly treated with intra carotid rSaK (0.1mg/kg, 0.2mg/kg, 0.4mg/kg) or 5% glucose. We observed angiographic efficacy and hemastatic function serially. RESULTS We find that the higher the rSaK dosage was, the higher the arterial recanalization were achieved. The highest dosage group decreased plasminogen significantly. CONCLUSIONS The rSaK can effectively dissolve trombus by intra arterial infusion, and the best dosage is 0.2mg/kg.
