雄激素及其拮抗剂对MCF-7乳腺癌细胞株雄激素受体表达的影响 被引量:1

摘要 研究药理浓度的睾丸酮对MCF-7乳腺癌细胞株雄激素受体表达的作用,以及雄激素拮抗剂氟他胺和羟基氟他胺对该作用的影响。方法应用荧光激活细胞分离器(FACS)检测雄激素受体阳性的细胞。结果10-9~10-6mol/L的睾丸酮使MCF-7乳腺癌细胞雄激素受体表达下调;10-6mol/L的氟他胺能显著拮抗睾丸酮下调雄激素受体表达的作用(P<0.01);羟基氟他胺更显著拮抗睾丸酮的作用,使雄激素受体表达显著增多,并呈剂量相关性(10-8mol/L时P<0.05;10-6mol/L时P<0.001)。结论MCF-7乳腺癌细胞株中雄激素受体存在自动调节,氟他胺和羟基氟他胺上调雄激素受体的表达。 PURPOSE To investigate the regulation of androgen receptor expression by pharmacological concentration of testosterone and the effect of flutamide or hydroxyflutamide. METHODS The percentage of androgen receptor positive cells were detected by fluorescence activated cell sorter (FACS). RESULTS 10 -9 ~10 -6 mol/L testosterone could downregulation the expression of androgen receptor in MCF-7 breast cancer cell line; 10 -6 mol/L flutamide significantly antagonized the effect of testosterone, so did hydroxyflutamide, which significantly up regulated the expression of androgen receptor, and the effect significantly related to the concentration (10 -8 mol/L, P <0.05; 10 -6 mol/L, P <0.001). CONCLUSIONS Androgen can autoregulate the expression of androgen receptor in MCF-7 breast cancer cell line. Flutamide and hydroxyflutamide up regulate the expression of androgen receptor.
