摘要
目的检测压力负荷增加大鼠血浆内皮素和降钙素基因相关肽的浓度,进一步揭示压力负荷增加致左心室肥厚的发病机制。方法采用腹主动脉狭窄法所致压力负荷增加大鼠左心室肥厚模型,观察左室质量指数,采用放射免疫分析法检测血浆内皮素和降钙素基因相关肽的浓度。结果①模型组血浆内皮素的浓度高于对照组(P<0.01),左心室质量指数模型组低于假手术组(P<0.05),与血浆内皮素的浓度呈正相关。②模型组血浆降钙素基因相关肽的浓度均低于对照组(P<0.01),与左心室质量指数呈负相关。③血浆内皮素/降钙素基因相关肽比值在假手术组接近<1,在模型组接近1,改变趋势与左心室质量指数呈正相关。结论血浆内皮素、降钙素基因相关肽的浓度比例失衡参与了压力负荷增加致左心室肥厚的发生,二者在高血压及左心室肥厚的发生、发展中具有不可忽视的作用。
Objective To observe the changes of plasma endothelin and calcitonin gene-related peptide (CGRP) in rats with pressure overload, and to study the possible mechanism of left ventricular remodeling. Methods Left ventricular mass index (LVMI) and plasma endothelin (ET) and calcitonin gene-related peptide were measured in the ral model with myocardial hypertrophy of congestiveheart failure and pressure overload by abdominal aortic constriction by radioimmunoassay. Results (1)The plasma endothelin levels in the model group were obviously higher than those of sham group (P〈0. 01 ), LVMI in model group was higher than that in sham operation group (P〈0.05). positively related to level of plasma ET. (2)The level of plasma CGRP in model group was lower significantly than that in sham operation group (P〈0.01), and negatively related to LVMI. (3)Plasma ET/CGRP ratio was less than 1 in normal control group obviously, close to 1 in model group, positively related to LVH. Conclusion The levels of plasma ET and CGRP were related to LVH. The mechanism of heart failure might be related with the disorganization of ET and CGRP levels in plasma so as the left vcntricular hypertrophy.
