摘要
目的:鉴定人肺腺癌肿瘤相关巨噬细胞(TAMs)的活化表型。方法:分别以CD68/巨噬细胞甘露糖受体(MMR)或CD68/诱导型一氧化氮合酶(i NOS)双阳性作为巨噬细胞发生替代性活化或经典活化的标记;收集40例人肺腺癌新鲜组织,双标免疫荧光染色和激光共聚焦显微镜观察TAMs表达CD68和MMR(或i NOS)。以IL-4处理人巨噬细胞,RT-PCR和Western-blot分别检测其表达MMR和i NOS的情况。结果:40例人肺腺癌全部同时表达CD68和MMR,其中CD68/MMR双阳性细胞占CD68阳性细胞的79.7%;仅有3例同时表达CD68和i NOS,CD68/i NOS双阳性细胞占CD68阳性细胞的12.4%。IL-4处理后,人巨噬细胞显著表达MMR而不表达i NOS。结论:人肺腺癌TAMs可能呈现出替代性活化的表型特征。
Objective: To identify the activated phenotypes of tumor-associated macrophages(TAMs) in human lung adenocarcinoma.Methods: Co-expressions of CD68/macrophage mannose receptor(MMR) and CD68/inducible nitric oxide synthase(iNOS) were used as the markers of alternatively activated macrophages(aaMphi) and classically activated macrophages(caMphi),respectively.The activated phenotypes of TAMs were detected by double-labelled immunofluorescence staining and observed by laser confocal microscopy in 40 cases of human lung adenocarcinoma.Then,MMR and iNOS expressions in human macrophages treated with IL-4 were detected by RT-PCR and Western-blot.Results: In human lung adenocarcinoma specimens,MMR/CD68 double-positive macrophages were found in all cases,but only 79.7% were CD68-positive TAMs.In contrast,iNOS/CD68 co-expression was detected in only three tumor specimens with a 12.4% expression in CD68-positive TAMs.In human macrophages treated with IL-4,a high level of MMR expression was found as...更多 well as negative iNOS expression.Conclusion: TAMs in human lung adenocarcinoma is proved to be one kind of aaMphi probably.
