摘要
目的:探讨不同剂量阿托伐他汀对大鼠脑出血(ICH)后脑组织水肿、神经细胞凋亡、血浆基质金属蛋白酶-9(MMP-9)水平的影响.方法:采用胶原酶注入法建立大鼠ICH模型,干湿重法测定各组脑组织含水量、电镜观察脑组织超微结构变化、ELISA法检测血浆MMP-9的水平.结果:阿托伐他汀可明显减少大鼠ICH后脑组织含水量,阻断神经元细胞凋亡,降低血浆MMP-9水平.血浆MMP-9水平与其脑组织含水量之间呈正的直线相关关系.结论:阿托伐他汀能明显减轻大鼠ICH后脑水肿,降低MMP-9引发的脑损伤,保护大脑神经元细胞.
AIM: To explore the effects of various doses of atorvastatin on the edema of brain tissue, the apoptosis of neurocytes and the level of matrix metalloproteinase-9 ( MMP-9 ) in plasma after intracerebral hemorrhage (ICH) in rats. METHODS: Making models with ICH by injecting collagenase into rats' putamen. The brain water content was determined with the wet and dry weight method, the uhrastructural changes of brain tissue were observed by electronmicro-scopy, the level of MMP-9 in plasma was determined with ELISA. RESULTS: Atorvastatin showed significant effect in reducing the brain water content, blocking the apoptosis of neurones of the ICH rats and decreasing the level of MMP-9 in plasma. There was a positive linear correlation between the level of MMP-9 in plasma and the brain water content. CONCLUSION: Atorvastatin can obviously relieve the brain edema, decrease the brain injury caused by MMP-9 and protect neurons of the ICH rats.
