摘要
目的:探讨微波诱导鼠成骨肉瘤细胞系UMR106细胞凋亡及其适宜作用剂量、作用时间.方法:以微波不同剂量和时间作用于鼠成骨肉瘤细胞系,采用形态学观察、MTT法、软琼脂集落形成实验、FCM等方法观察微波诱导成骨肉瘤细胞的凋亡.结果:不同剂量的微波照射大鼠成骨肉瘤细胞后细胞形态发生了改变,随照射剂量的增高,细胞逐渐脱离基质;成骨肉瘤细胞在不同剂量的微波进行相同时间(1h)照射下存活率均显著下降,并呈剂量依赖性;微波作用于UMR106细胞的IC50为100W/m2微波辐射下不同时间后,细胞的存活率显著下降,呈时间依赖性;微波辐射下大鼠成骨肉瘤细胞出现G1期阻滞.结论:微波辐射可诱导大鼠成骨肉瘤细胞凋亡,其最佳作用时间和剂量为1h和100W/m2.
AIM:To elucidate the appropriate dose and time of microwave to induce the apoptosis of UMR 106 osteogenic sarcoma cells.METHODS:Treat the osteogenic sarcoma cells with microwave of different dose and time and use morphology detection,MTT assay,soft agar formation and FCM analysis to examine the apoptosis level.RESULTS:Different dose of microwave caused the morphology change of osteogenic sarcoma cells.With increased dose,more and more cells detached from basal layer.The survival rate of UMR 106 cells decreased a lot following the radiation of microwave with 1 h treatment and demonstrated the dose-dependent trend in MTT assay.The IC50 of microwave radiation on UMR 106 cells was 100 W/m2.FCM analysis showed the G1 arrest of cell cycle in UMR 106 cells under the treatment of microwave radiation.CONCLUSION:Microwave radiation can induce the apoptosis of UMR 106 osteogenic sarcoma cells and the opitimized time and dose were 1 h and 100 W/m2 respectively.
