Establishment of transplantation tumor models for Walker-256 rats and H 22 mice and discussion on their applications

摘要 目的研究大鼠Walker-256及小鼠H22两种肿瘤模型的成瘤性和稳定性,并探讨其应用。方法建立大鼠Walker-256及小鼠H22两种皮下肿瘤模型,1周后观察其出瘤率和肿瘤大小,观察并绘制肿瘤生长曲线,实验结束后进行病理学检测。结果1周后两种鼠肿瘤模型出瘤率分别为98.57%、97.14%(P>0.05);肿瘤直径分别为(9.61±2.90)mm、(8.25±1.89)mm(P>0.05);大鼠Walker-256模型有肿瘤自然消退现象,而小鼠H22模型肿瘤生长稳定且少数出现肺转移;病理学检测支持上述现象。结论小鼠H22皮下肿瘤模型较大鼠Walker-256模型稳定,可用于抗肿瘤疗效评价。 Objective To establish two transplantation tumor models for Walker-256 rats and H 22 mice, and to discuss their applications. Methods Subcutaneous transplantation tumor-bearing models for Walker- 256 rats and H 22 mice were established, respectively. The positive rate of inoculation and tumor size were observed after one week. The volume of tumor was measured every two days to construct tumor growth curve. Pathological features of tumor were observed after the experiment was finished. Results The positive rate of inoculation was 98.57% in Walker-256 rats and 97.14% in H 22 mice (P 〉 0.05). The tumor size was (9.61 ± 2.90) mm in Walker-256 rats and (8.25± 1.89) mm in H 22 mice ( P 〉 0.05). During the follow-up measurements, it was found that the tumors in Walker-256 rats reduced in size, some of them even disappeared completely, while the tumors in group H 22 mice grew steadily. Lung metastasis was observed in some mice. The pathological findings supported these results. Conclusion Subcutaneous transplantation tumor-bearing model for H 22 mice is more stable than the model for Walker-256 rats and is suitable for the effect evaluation on inhibiting tumor.
