摘要
目的:观察参附注射液对Wistar大鼠肝脏部分缺血再灌注损伤的保护作用。方法:以Wistar大鼠复制肝脏部分缺血再灌注损伤模型,动物随机分为参附注射液组、对照组(生理盐水注射)及假手术组。动物缺血90min再灌4h采集标本,检测门静脉血清TNF-α、IL-10及血浆ALT水平,肝叶组织常规固定切片HE染色,光镜下观察。结果:肝脏缺血90min再灌注4h后参附注射液组的鼠血清IL-10水平比对照组增高,而血清TNF-α及ALT水平明显降低。结论:参附注射液可以通过提高IL-10、抑制TNF-α减轻肝脏缺血再灌注损伤。
Objective: to investigate the protective effect of Shenfu Injection in mouse partial live ischemia/reperfusion injury in Wistar mice.Methods: The model of the mouse partial live ischemia/ reperfusion injury was established. All the animals were divided randomLy into 3 groups: Shenfu group, vehicle (0.9% sodium chloride solution) group and sham group. Mice were sacrificed after4h to analyze the acute phase of liver IRI. The level of TNF-α , IL-10 and ALT in portal vein were measured in all groups. Pathological sections of liver (HE staining) were used for histological analysis. Resu/ts: After restoration of blood supply, The IL-10 of portal vein was up-regulated markedly in Shenfu group compared to vehicle group, contrary, the level of TNF-α and ALT of portal vein was down-regulated markedly in Shenfu group compared to vehicle group at each point of time in mice partial liver UR injury model. Conclusion: Shenfu Injection raise IL-10 and inhibition the TNF- α mediated inherent immune response and it may play crucial role in lessen the liver UR injury.
