摘要
目的:建立浙江省沙门菌PFGE指纹图谱资料库,为查明沙门菌食源性疾病污染源,确定各病例之间的病原学关系,及时处理大范围内呈散发特征的沙门菌事件提供科学依据。方法:收集、提取全省历年来在各类食品和腹泻病人中分离的209株沙门菌的DNA,用限制性内切酶Xbal切割,脉冲场凝胶电泳(PFGE)技术建立PFGE指纹图谱,用BioNu-merics V5.1软件UPGMA方法进行聚类分析。结果:209株沙门菌分成10个基因型别160种PFGE基因指纹图谱。其中67株德尔卑沙门菌可分为6个型别35种PFGE指纹图谱,主要集中在1个PFGE指纹图谱型别中。14株肠炎沙门菌分属3个型别8种PFGE指纹图谱,相似系数在0.800-1.000之间。鼠伤寒沙门菌间的PFGE指纹图谱差异较大。结论:PFGE指纹图谱与血清学型别无相关性,该方法在研究沙门菌分子流行病学、确定各病例之间的病原学关系效果良好。
Objective:To establish the PFGE fingerprint data bank of Salmonella in Zhejiang province and provide scientific evidences to pollution source survey to find out the relationship of patients and resolve the extensive sporadic cases.Methods:DNA collected from 209 strains which were separated from food and diarrhea patients in province through years,digested DNA by XbaI and established the fingerprint data bank,clustering analysis by BioNumerics V 5.1.Results:209 strains of Salmonella were classified as 10 gene types and 160 PFGE fingerprint types.67 strains of Salmonella Derby were classified as 6 gene types and 35 PFGE fingerprint types,most of them were the same gene type.14 strains of Salmonella Enteritidis were classified as 3 gene types and 8 PFGE fingerprint types,the similarity coefficient was between 0.800 to 1.000.PFGE fingerprint of Salmonella Typhimurium were variant.Conclusion:PFGE fingerprint types are not relatively with setotypes,the results of PFGE are good at molecular epidemiology analysis and finding out the relationship between cases.
