摘要
目的:研究宁波市三带喙库蚊(Culex tritaeniorhynchus)携带乙脑病毒的基因型别。方法:采用Real-Time PCR方法,检测2007年6月至9月捕获到的91批三带喙库蚊乙脑核酸。利用C6/36细胞培养方法从阳性标本中分离乙脑病毒。扩增毒株E基因并测序,将结果与GenBank中其他乙脑病毒代表株序列进行比较,用计算机软件分析结果。结果:共有6批三带喙库蚊乙脑Real-Time PCR检测结果成阳性,从中分离到2株乙脑病毒,全为GⅠ基因型。2株乙脑毒株与疫苗株SA14-14-2 E基因氨基酸同源性分别为96%和97%。结论:宁波市三带喙库蚊携带GⅠ型乙脑病毒。
Objective:To study molecular epidemiology of Japanese encephalitis virus(JEV) from culex tritaeniorhynchus in Ningbo.Methods:Totally 5400 mosquitoes have been collected from June to September in 2007.JEV in mosquitos were identified by Real-Time PCR,and positive specimens were inoculated to C6/36 monolayer cells to isolate virus.The nucleotide fragment in E gene was amplified for sequencing,and results were analyzed by BIOEDIT and MEGA software.Results:Two strains of Japanese encephalitis virus were isolated from mosquito and both were identified as genotype I.Sequence analysis showed that the amino acid homeology between the two strains of encephalitis virus and the strain of vaccine SA 14-14-2 was 96% and 97%,respectively.Conclusion:Culex Tritaeniorhynchus in Ningbo has genotype I JEV.
