HPLC-ELSD法测定乳粉及米粉中功能性低聚果糖 被引量:7

摘要 目的:建立简便、准确的测定乳粉及米粉中低聚果糖(蔗果三糖、蔗果四糖、蔗果五糖)的高效液相色谱蒸发光散射检测方法。方法:试样除去蛋白,离心、浓缩、定容、过膜后,用液相色谱分析,用聚合相NH2柱分离,蒸发光散射检测器测定,外标法定量。结果:低聚果糖的测定范围为:0.05-1.0 mg/g、检出限为0.01 mg/g,回收率在95%-105%之间,测定的相对标准偏差:Res(%)〈±5。结论:该方法快速、灵敏,适用于婴幼儿食品中添加的低聚果糖含量的测定分析。 Objective:To establish a simple and precise method for determination of fructooligosaccharides(1-Kestose,Nystose and 1-Fructofuranosylnystose) in milk and rice flour by HPLC-ELSD.Methods:Samples were deproteinized,centrifuged,concentrated,fixed volume and filtered,then were separation and determined by HPLC with NH2 column connected with ELSD detector.Results:The scope of determination ranged from 0.05 mg/g to 1.0 mg/g,the detection limit was 0.02 mg/g,the recovery was 95 ~105%,the RSD(%)〈±5.Conclusion:The method is sensitive,and suitable for rapid determination and quality control of fructooligosaccharides in food for infants.
