摘要
目的:建立快速检测四环素残留的胶体金免疫层析检测方法。方法:采用免疫竞争法,将制备的胶体金颗粒与抗四环素单克隆抗体结合形成抗四环素单克隆抗体-金标复合物标记在胶体金结合垫上。并将四环素人工抗原包被在硝酸纤维素膜上作为检测线(T线)。其与待测样品中相应四环素类药物竞争结合胶体金标记的抗四环素类单克隆抗体,并能以颜色直观显示检测的定性结果。结果:检测肉类试样时,灵敏度最低值可达到10 ng/ml,只需3~5 min,与其它兽药无交叉反应。结论:该方法灵敏度高、特异性强、稳定性好,可作为四环素类残留批量检测的筛选方法。
Objective:To establish the method of colIloidal gold immunochromatographic assay for detecting tetracyclines residues.Methods:Nanocolloidal gold particles were prepared and labeled to an anti-tetracyclines monoclonal antibody.Tetracyclines was conjugated to bovine serum a1bumin(BSA)and dispersed on a nitrocellulose membrane to be the test 1ine(T).The more analyte present in the sample,the more effectively it would compete with the Tetracyclines-BSA for binding to the limited amount of gold-labeled anti-tetracyclines monoclonal antibody.The presence or absence of a colored band on the test line indicated a negative or positive result.Results:When measuring the meat sample spiked with tetracyclines,the minimum detection concentration can reach 10 ng/ml.The major advantage of the one-step strip test is that the test can be completed within 5 minutes.No cross-reaction occurred between tetracyclines and other veterinary drugs.Conclusion:The method for tetracyclines is of high sensitivity,specificity and good stability and it could be widely used to detect the tetracycline residues.
