摘要
柑橘植物组织中富含多糖、酚类、单宁、色素等物质,它们的存在严重干扰了柑橘黄龙病PCR检测的准确性。因此有必要筛选出一种能得到纯度高、结构完整的柑橘总DNA(含黄龙病病原DNA),且操作简便的提取方法。本研究对比试验了3种柑橘总DNA提取方法,方法1采用Sandrine等人的提取法;方法2采用CTAB法;方法3采用试剂盒法。实验研究表明,方法2提取柑橘黄龙病病原DNA的PCR检测准确率最高。
The PCR test of citrus Hulongbing was affected by some substance in citrus including polysaccharides,Phenolic,tannic acid,pigment and so on.It was necessary to establish a simple method to extract high purity and integrated construction citrus DNA.Three method were used in this experiment.There were Sandrine method,CTAB method,and Reagent box method.The results showed in these methods,CTAB was the best one in the three.
