摘要
目的探讨大鼠星形胶质细胞的最优体外培养方法与培养条件。方法采用差异贴壁等手段进行组织原代培养,胰蛋白酶消化传代,并应用相差显微镜和即用型SP-9001免疫组化试剂盒对细胞进行形态学和免疫组化鉴定。结果镜下培养细胞生长多呈多形性、扁平状,胞体较大,胞突较多较长,细胞核呈圆形或卵圆形,常偏于胞体的一侧,免疫组化染色显示细胞浆为棕褐色,细胞核为蓝色,呈阳性表达,星形胶质细胞的纯度在95%以上。结论本法可获纯度高、结构和功能良好的大鼠星形胶质细胞,可作为体外的细胞模型为研究细胞因子对其功能的调控作用奠定实验基础。
Objective To study the optimum culture method and condition of astrocyte in vitro. Methods The cells were cultured in the method of different adherent tlme,subculture with trypsinization. They were detected by microscopy with the phase contrast microscope. The ready-to-use immunohistochemistry SP-9001 kit was used to identify the cells by means of immunohistochemistry, Results Viewing through the phase contrast microscope,the cells were flattened and the body was big,with many and long cytoplasmicprocess. The cell nudears were round or ovoid in shape on one side of its body. Immunohistochemical staining showed that the cytoplasm was dark brown and the cell nuelears were blue, which was positive result.It also demonstrated that the purity of the astroeyte was higher than 95%. Conclusion In this method,the astrceyte of rats were high purity,and the structure and function of them were good. The coils can he used to study the function and centrol the cell factors as cell model in vitro.
