淫羊藿女贞子配伍对哮喘大鼠模型Th1/Th2调节作用的研究

Experimental study of Yinyanghuo(Herba Epimedii),Nuzhenzi(Fructus Ligustrilucidi) on Disequilibrium of Th1/Th2 in Asthmatic Rat

目的:研究淫羊藿女贞子配伍对哮喘大鼠模型Th1/Th2失衡的免疫调节作用。方法:采用OVA(ovalbumin,卵蛋白)致敏、激发复制大鼠慢性哮喘模型,以阿斯美胶囊为对照药,分别通过放免法及酶免法检测血及BALF(Bronchoalveolar lavage fluid,支气管肺泡灌洗液)中IL-4及IFN-γ的含量、免疫组化法检测肺组织IL-4和IFN-γ蛋白表达,RT-PCR技术检测肺组织T-bet和GATA-3 mRNA的表达,探讨淫羊藿女贞子对大鼠哮喘模型Th1/Th2调节途径的影响作用。结果:淫羊藿女贞子能显著抑制BALFIL-4含量(P<0.01),并降低血及BALF中IL-4/IFN-γ比值(P<0.05);对肺组织IL-4蛋白表达的Area有降低作用(P<0.05),并能抑制IL-4/IFN-γ Area的比值增高(P<0.01);上调肺组织T-bet mRNA表达,下调GATA-3 mRNA表达(P均<0.05)。结论:淫羊藿女贞子对哮喘大鼠Th1/Th2失衡有一定的调节作用。

Objective： To study the immune adjustment effects of the compatibility of Yinyanghuo and Nuzhenzi on the imbalance of Th1Th2 in asthmatics. Methods：Rat asthma model was duplicated by OVA through sensitizing and chal- lenging. Asimei Capsule was used as positive control medicine. By investigating levels of IL -4 and IFN - γ f blood and BALF, protein expression of IL - 4 and IFN - γ , T - bet and GATA - 3 mRNA expression in lung tissue, to explore the adjustment on Thl/Th2. Results： The compatibility of Yinyanghuo and Nuzhenzi could inhibit IL- 4 content in BALF （P 〈 O. O1 ）, and lower the ratio of IL - 4 to IFN - γin blood and BALF （ all P 〈 0.05 ）, reduce the area of protein expres- sion of IL - 4 in lung tissue （ P 〈 0.05）, increase T - bet mRNA expression, and decrease GATA - 3 mRNA expression in lung tissue （ all P 〈 0.05 ）. Conclusion ： The compatibility of Yinyanghuo and Nuzhenzi has the effect on the imbalance of Thl/Th2 in asthmatic rats.
