摘要
目的:观察Chemerin基因mRNA在小鼠体内脏器及在3T3-L1脂肪前体细胞诱导分化过程中的表达情况。方法:体外培养3T3-L1脂肪前体细胞,诱导分化过程中收集9个不同时间点的细胞;同时取正常小鼠7种脏器,一并提取脏器组织和细胞中总RNA,半定量RT-PCR检测其中Chemerin基因mRNA表达水平。结果:Chemerin在脂肪组织、肝脏和肾脏为高;在3T3-L分化的过程中Chemerin基因表达量显著上调。结论:Chemerin是一种新的脂肪因子,参与脂肪细胞的分化,可能参与肥胖等代谢性疾病的发生。
Objective : To observe the change of Chemerin gene mRNA expression in various tissues of mice and in 3T3-L1 preadipoeyte differentiation. Methods: Culture of 3T3-L1 cells and induction of differentiation were performed in vitro. Total RNA was extracted from seven kinds of tissues of mice and the differential cells at nine time points. Semi-quantitative RT-PCR was performed in order to measure levels of Chemerin mRNA. Results: Chemerin mRNA was expressed most highly in adipose tissue, liver and kidney. Chemerin mRNA expression dramatically increased during the process of differentiation of 3T3-L1 cells. Conclusions: Chemerin is a novel adipokine that participates in preadipocyte differentiation which indicates that it may play a key role in the development of obesity and other metabolic diseases.
