摘要
目的了解糖尿病患者外周血Obestatin、Ghrelin水平变化,探讨Obestatin、Ghrelin在糖尿病发病中的作用。方法采用酶联免疫吸附分析方法(ELISA)测定42例糖尿病患者和20例正常健康人血浆中Obestatin、Ghrelin水平,放射免疫法(RIA)测定血浆中空腹胰岛素水平(FPI)。同时检测身高、体重、血压,空腹血糖(FPG)、血三酰甘油(TG)、血胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)。结果糖尿病患者血中Obestatin、Ghrelin水平均明显低于健康对照组[(2.06±0.66)vs(3.01±1.09)ng/ml和(3.05±0.95)vs(3.84±1.39)ng/ml,均P<0.05]。G/O比值无明显变化(P>0.05)。肥胖糖尿病组Obestatin、Ghrelin水平明显低于非肥胖糖尿病组[(1.83±0.49)vs(2.30±0.75)和(2.77±1.01)vs(3.37±0.78)ng/ml,均P<0.05]。相关分析表明Ghrelin、Obestatin均与BMI、HOMA-IR、FPI、FPG负相关(均P<0.05)。结论Obestatin、Ghrelin水平在糖尿病的发生中起重要作用。
Objective To investigate the levels of circulating Obestatin and Ghrelin in Diabetes mellitus.Methods The levels of plasma Obestatin and Ghrelin in 42 patients with diabetes mellitus and 20 normal control were measured by enzyme linked immunosorbent assay(ELISA) and fasting plasma insulin(FPI) concentrations were measured by radioimmunoassay(RIA).Body height,body weight,blood pressure were measured,fasting plasma glucose(FPG),circulating triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C) were measured.Results The levels of plasma Obestatin and Ghrelin in diabetes mellitus group were decreased significantly than those in the normal control group[(2.06±0.66) vs(3.01±1.09) ng/ml and(3.05±0.95) vs(3.84±1.39) ng/ml,all P〈0.05],while there were no significant differences of G/O ratios between two groups(P〉0.05).The levels of plasma Obestatin and Ghrelin in diabetes mellitus obesity group were decreased significantly than those in the Diabetes mellitus with normal body mass index(BMI)[(1.83±0.49) vs(2.30±0.75)and(2.77±1.01) vs(3.37±0.78) ng/ml,all P〈0.05].There were significant negative correlations respectively between the levels of Obestatin、Ghrelin and BMI,HOMA-IR,FPI,FPG(all P〈0.05).Conclusion The concentrations of circulating Obestatin and Ghrelin may play an important role in the development of diabetes mellitus.
