摘要
目的研究红景天苷对局灶性脑缺血/再灌注损伤后突触结构和数密度的变化。方法采用线栓法制造大鼠大脑局灶缺血(2h)/再灌注模型(I/R模型),于灌注后1d、3d、7d、14d时间点断头取脑,电镜观察突触结构和数密度的变化。结果I/R模型组数目减少,7d降至最低,14d突触数目回升;突触结构随再灌注时间延长损伤加重,14d有所恢复;红景天组突触损害程度减轻,突触数目增加(P<0.05),突触数密度恢复的时间提早至7d。结论红景天苷可以减轻脑缺血再灌注后突触的损伤,易化突触可塑性。
Objective To explore the effects of salidroside on synapse ultramicrostructure following focal cerebral ischemia/ reperfusion injury. Methods The model of the middle cerebral artery occlusion(2 h)/reperfusion in rats was prepared by using the intraluminal filament occlusion, The changes of synapse structure and density were observed by electron microscope at 1 d, 3 d, 7 d, 14 d after reperfusion. Results I/R control group The synapse densities were decreased after reperfusion, reached its lowest level at 3d. It began to elevate at 14 d. The synapse ultrastructure showed advanced destruction, and improved on 14 d. In the Salidroside-treat group the damage of the synapse ultrastructure decreased while the number of synapse increased (P〈0.05). The recovery of synapse densities were shift to an earlier time at 7 d. Conclusion Salidroside can relieve the synapse damage following focal cerebral ischemia/reperfusion injury, suggesting that rhodioside can ease the synapse-plasticity.
