摘要
目的:研究5-氨基酮戊酸光动力疗法(ALA-PDT)治疗豚鼠耳背部寻常型银屑病模型的可行性和作用机制。方法:采用20%ALA溶液局部封包豚鼠耳背部银屑病模型皮损4h,再用635nm半导体激光照射,能量密度12J/cm^2。分设模型对照组、单纯照光组、单纯ALA组、一次ALA-PDT治疗组、二次ALA-PDT治疗组,治疗后于第7d、14d、21d、28d进行组织病理学和增殖细胞核抗原免疫组化观察疗效。结果:ALA-PDT能够治愈豚鼠耳背部寻常型银屑病模型的病变;二次ALA-PDT治疗组疗效优于一次ALA-PDT治疗组;结论:ALA-PDT疗法能抑制豚鼠耳背部寻常型银屑病模型的表皮细胞异常增殖,并能消除局部组织炎性反应,其疗效与治疗次数正相关。
Objective: This research is devoted to investigate the feasibility and the mechanism of action on 5-Aminolevulinic Acid Photodynamic Therapy (ALA-PDT) treating psoriasis vulgaris model of guinea pigs' ear back. Method: This research adopted 20% ALA solution wet packing lesions for four hours. Then, it used irradiation with the semiconductor laser of 635nm wavelength and energy density 12J/cm^2. This experiment was divided into model control group, simple irradiation group, simple ALA group, once ALA-PDT treatment group and twice ALA-PDT treatment group. The observation of each group's histopathological and Proliferating Cell Nuclear Antigen immunohistochemical efficacy was conducted at day 7, 14, 21 and 28. Resuit: ALA-PDT had a significant curative effect in treating psoriasis vulgaris model of guinea pigs' ear back. The curative effect of the twice ALA-PDT treatment group was better than the once ALA-PDT treatment group. Conclusion: ALA-PDT can inhibit the epidermal cell proliferation of psoriasis vulgaris model of guinea pigs' ear back. It can also eliminate the inflammatory reaction of the local tissue. There is a positive correlation between the curative effect and treatment times.
