肾上腺髓质素(13-52)对Wistar大鼠损伤心肌保护作用的初步研究

摘要 目的观察肾上腺髓质素(AM13-52)对Wistar大鼠心肌损伤和坏死指标的作用。方法将250~300 g雄性健康Wistar大鼠56只,分为对照组、异丙肾上腺素(ISOP)心肌坏死组、ISOP+AM组和ISOP+AM+左旋硝基精氨酸(L-NNA)组,分别观察心肌保护指标(心肌cGMP)、心肌损伤指标(心脏/体质量指数、心肌丙二醛(MDA)含量、心肌Ca2+含量)、血浆内皮素(ET-ir)以及NO活性指标(cGMP)水平的动态变化。结果4组各指标比较差异均有统计学意义。ISOP组与对照组比较,血浆ET-ir、乳酸脱氢酶(LDH)、MDA、心脏/体质量指数、心肌MDA含量及Ca2+含量均有所增高,心肌cGMP和血管cGMP含量均明显降低,差异均有统计学意义(P<0.01);ISOP+AM组与ISOP组比较,血浆ET-ir、LDH、MDA心脏/体质量指数、心肌MDA含量及Ca2+含量明显下降,心肌cGMP和血管cGMP含量明显增高差异均有统计学意义(P<0.01);ISOP+AM+LNNA组与ISOP+AM组比较,血浆ET-ir、LDH、MDA心脏/体质量指数、心肌MDA含量及Ca2+含量又有所增加,心肌和血管cGMP水平下降,各指标比较,差异均有统计学意义(P<0.01)。结论AM对损伤坏死的心肌有重要的保护作用,与NO的允许作用直接相关,可能通过NO介导或者通过改善NO缺陷进而拟制对心肌损伤的保护效应。 Objective To observe that AM(13-52)plays a role in injuring and protecting idexes of myocardium in wister rats.Methods To take 56 Wistar rats which weight 250~300g into control group,ISOP myocarclial necrosis group,ISOP+ AM group,ISOP+ AM+ L-nityl-arginine(L-NNA)group.Respectively to observe moving vary of cardiac muscule protective idexes(cardiac muscule cGMP),myocarclial injunry idexes(heart/body quality ratio,mail delivery agent(MDA)content and Ca^2+ content of cardiac muscule),plasm endothelin and cGMP,a NO active idex.Results In 4 groups,each difference between the indicators are statistically significant.ISOP group compared with control group,plasm ET-ir,Lactate dehydrogenase(LDH),MDA,heart/body ratio,MDA level,content of Ca^2+ of cardiac muscle are all increased,the cGMP level of cardiac muscle and,the cGMP level of blood vessel are all reduced,all diference had statistics significance(P〈0.01).ISOP+AM group compared with ISOP group,plasm ET-ir,Lactate dehydrogenase(LDH),MDA,heart/body ratio,MDA level,content of Ca^2+ of cardiac muscle are all reduced,the cGMP level of cardiac muscle and,the cGMP level of blood vessel are all obvious increased,all difference had statistics significance(P〈0.01).As compared with ISOP+AM group,above effects in with ISOP+AM group may be obviously reversed in ISOP+AM+L-NNA group(all idexes,P〈0.01).Conclusion AM plays an important role in protecting myocarclial necrosis and injury.It′s mechanism is directly relativity with NO permissive action,it is likely transduced by NO,or by mechaism improved NO defect,the effect of myocardial preservation is carried out.
