摘要
目的:研究雷公藤多苷对苯巴比妥大鼠P450含量及血药浓度的影响,指导临床安全、合理使用雷公藤。方法:选用苯巴比妥诱导的大鼠作为对照组,合用雷公藤多苷作为实验组,再设空白组,分别测定各组各时间点的P450含量;分别测定对照组和实验组各点苯巴比妥的血药浓度。结果:合用组对肝药酶呈现较强的诱导作用并符合P450含量自身的节律变化,合用雷公藤后对苯巴比妥的诱导作用有明显的促进作用。雷公藤使苯巴比妥的达峰时间前移。结论:代谢雷公藤的P450酶和苯巴比妥所诱导的P450酶之间存在着必然的联系。
Objective: To investigate effects of Tripterygium Glycoside(TG) on cytochrome P450 serum concentration in phenobarbital-treated rat. Methods: Test rats were separately divided into three groups: phenobarbital-treated rat control group;combined TG experimental group and blank group, phenobarbital and P450 concentration were measured at different time points in each group. Results Combination group showed strong induction and accorded with its own rhythm of change, it expedited induction of Phenobarbital obviously. phenobarbital Tmax was moved forward by Tripterygium Glycoside. Conclusion: There were inevitably link between Tripterygium Glycoside P450 metabolic enzymes and induced P450 enzyme by Phenobarbital.
