摘要 目的:探讨Caspase-3及Bcl-2的异常表达与骨肉瘤发生、发展的关系。方法:应用免疫组织化学方法检测骨肉瘤、骨软骨瘤中Caspase-3和Bcl-2的表达,并结合临床资料对结果进行统计学分析。结果:在41例骨肉瘤患者中,Caspase-3阳性19例,阳性率为46.3%,低于骨软骨瘤中Caspase-3的表达;其中G1期骨肉瘤表达明显高于G2期(P<0.05);Bcl-2阳性表达14例,阳性率为34.1%,显著高于骨软骨瘤;其中G1期组的表达明显高于G2期组(P<0.001)。14例Bcl-2阳性的骨肉瘤病例中,Caspase-3阳性13例,两者阳性比率为92.9%;27例Bcl-2阴性的病例中,Caspase-3阴性22例,两者阴性比率为81.5%。两者有很好的相关性(P<0.001)。结论:Bcl-2对细胞凋亡的抑制是骨肉瘤的早期分子事件,同骨肉瘤的发生关系密切,Caspase-3表达下调同骨肉瘤的发生、发展有关,在骨肉瘤中细胞凋亡的抑制是两者相互作用的结果。 Objective: To explore the relationship between the abnormal expression of Bcl-2 and Caspase-3 and the oc currencc as well as the development of osteosarcoma (Os). Methods: Immunohistochemistry was used to detected the expression of Bcl-2 and Caspase-3 in osteosarcoma and osteochondroma (Oc), and performed statistical analysis of the results combined with the clinical data. Results: For the 41 cases of Os, 19 cases (46.3%) showed Caspase-3 expression, which was lower than that of Oc. The expression of G1 stage Os was significantly higher than that of G2 stage Os (P〈0.05). 14 cases (34.1%) showed Bcl-2 expression, significantly lower than that of Oc. The expression of G1 stage was significantly higher than that of G2 stage (P〈0.01). For the 14 cases of Os that expressed normal Bcl-2 level, 13 cases showed Caspasc-3 ex- pression, their positive rate was 92.9%. For the 27 cases of Os that expressed decreased Bcl-2 level, 22 cases showed decreased Caspase-3 expression, their negative rate was 81.5%. The correlation was found between Caspasc-3 and Bcl-2 (P〈 0.001). Conclusion: Bcl-2 restricts programmed cell death on early stage in Os, and closely associated with the occurrence of Os. The clown-regulation of Caspase-3 expression is a common event in Os. So it is associated with occurrence and development of Os. Caspase-3 and Bcl-2 were co-expressed in Os. This suggest that, in Os, the pregrammed cell death restriction is a result of their mutual action.
