摘要
目的研究黄芪皂甙(saponins of astrag-alus,SA)对豚鼠心室肌细胞动作电位(AP)的作用。方法实验采用标准玻璃微电极细胞内记录心室肌细胞动作电位(AP)。结果应用SA20mg/L后心室肌细胞动作电位的幅度从给药前的(69.31±6.07)mV降低到(57.37±3.68)mV(n=6,P<0.05);动作电位时程(APD90)从给药前的(263.38±16.36)ms延长到(284.33±21.21)ms(n=6,P<0.05)。应用SA40mg/L后心室肌动作电位的幅度从给药前的(71.37±56.34)mV降低到(47.23±8.67)mV(n=6,P<0.05);动作电位时程(APD90)的从给药前的(238.31±13.28)ms延长到(314.64±27.36)ms(n=6,P<0.05)。结论SA可以降低心室肌动作电位幅值延长动作电位时程,并且有一定的剂量依赖性。
Objective To study the effects of saponins of astrag-alus (SA) on the action potential and the delayed rectifier potassium channel in ventricular myocardium of guinea pig Method: The action potential was recorded in guinea pig ventricular myocyte by using of standard microelectrodes technique.Results The action potential amplitude (APA) of ventricular myocardium was decreased from (69.3 1±6.07)mV to (57.37±3.68)mV (n=6,P〈0.05), the duration (APD90) of ventricular myocardium was expanded from (263.38±16.36)ms to (284.33±21.21)ms (n=6,P〈0.05) after application of SA dosage of 20 mg/L.The APA of ventricular myocardium was decreased from (71.37±56.34)mV to (47.23±8.67) mV (n=6,P〈0.05) and the APD90 of ventricular myocardium was prolonged from (238.31±13.28)ms to (314.64±27.36)ms (n=6,P〈0.05) after application of SA dosage of 40 mg/L. Conclusion It has been suggested that SA decrease action potential amplitude, prolong action potential duration in guinea pig remarkably with concentration-dependence.
