奥美沙坦对自发性高血压大鼠延髓腹外侧区AT1受体mRNA表达的影响

Effect of Olmesartan on the Expression of AT1 Receptor in Rostral Ventrolater Medulla of the Spontaneously Hypertensive Rat

摘要 目的探求奥美沙坦(olmesartan)对自发性高血压大鼠(SHR)延髓腹外侧头端区(RVLM)血管紧张素Ⅱ1型(AT1)受体mRNA表达的影响。方法实验大鼠分作SHR对照组(正常饮水)、SHR-olme组[奥美沙坦,30mg(/kg·d)]和WKY组(正常饮水)。4周后,断头取材,采用RT-PCR方法检测RVLM内AT1受体mRNA的水平,并进行光密度测定。结果给药1周后奥美沙坦组血压均能够达标。SHR-olme组RVLM内AT1受体mRNA平均光密度(MOD)明显低于SHR组[(0.94±0.41)vs(2.41±0.37),P<0.01],但高于WKY组(0.81±0.22,P<0.05)。结论奥美沙坦能够显著减少SHR的RVLM组织中AT1受体mRNA表达。 Objective To study the effect of olmesartan on the expression of AT1 receptor mRNA in rostral ventrolater medulla(RVLM) of the spontaneously hypertensive rat (SHR). Methods Rats were divided into SHR conical group (normal drinking), SHR-Olme group [ (ohnesartan,30 mg/(kg·d)] ,and Wistar-Kyoto rats (WKY) group (normal drinking ) . Four weeks later,these rats were decapitated to death and got the brain tissue. AT1 receptor mRNA level in RVLM was measured by RT-PCR and analyzed with optic density. Results The blood pressure of all SHR-olme rats reached normal level after administration of olmesartan for a week. The mean optic density (MOD) of A T1 receptor mRNA in RVLM of SHR-olme was much reduced compared with SHR group [ ( 0.94 ±0.41 ) vs (2.41±0.37 ), P 〈 0.01 ], but higher than that of WKY group(0.81±0.22, P 〈 0.05 ). Conclusion Olmesartan could singificantly reduce the leve of AT1 receptor mRNA in RVLM of the SHR.
