缺氧诱导因子-1α在喉鳞状细胞癌中的表达及与微血管密度的关系 被引量:1

Expression of Hypoxia Inducible Factor-1α in Human Laryngeal Squamous Cell Carcinoma and its Relationship with Microvessel Density

摘要 目的分析缺氧诱导因子-1α(HIF-1α)在喉鳞状细胞癌中的表达及与微血管密度(MVD)的关系,探讨其与喉癌生物学行为及预后的关系。方法采用免疫组化法检测50例喉癌标本中HIF-1α的表达,并计数MVD,观察HIF-1α、MVD与喉癌各种病理因素之间的关系。结果50例喉癌标本中39例HIF-1α表达阳性,平均MVD为(28.0±11.3),与对照组比较差异有统计学意义(P<0.05)。T分期晚期患者的HIF-1α阳性表达率为89.7%,MVD为(32.5±11.5),明显高于早期患者的61.9%和(21.7±7.7);淋巴结转移组中HIF-1α的阳性表达(94.4%)及MVD(37.1±8.7)明显高于无淋巴结转移组中的68.8%和(22.9±9.3),二者差异有统计学意义(P<0.05)。HIF-1α阳性组的MVD明显高于阴性组;随着HIF-1α表达的增加,MVD逐渐增高,呈正相关(r=0.901,P<0.05)。结论HIF-1α在喉鳞状细胞癌中的表达与肿瘤的生物学行为及预后有关,提示其在喉癌的浸润和转移中起了重要作用。 Objective In order to evaluate the role of hypoxia inducible factor-1α (HIF-1α) protein in human laryngeal squamous cell carcinoma and its relationship with micmvessel density (MVD). Methods The expression of HIF-1α was detected in 50 laryngeal squamous cell carcinomas and 10 normal laryngeal tissues using inmunohistochemical technique. The MVD was assessed simultaneously. The relationship between them was analyzed. Results The expression of HIF-1α (78%,39/50) and the average MVD (28.0±11.3) in laryngeal squamous cell carcinomas were significantly higher than the normal control (P 〈 0.05). HIF-1α expression (89.7%) and MVD (32.5± 11.5) in the tumors of T3±T4 stage was significantly higher than that of T1±T2 stage (61.9% and 21.7±7.7) (P 〈 0.05). HIF-1α expression (94.4%) and MVD (37.1±8.7) in the tumors with lymphnode metastasis were significantly higher than that without lymphnode metastasis (68.8% and 22.9±9.3) (P〈 0.05). MVD in HIF-1α positive group (31.1±10.1) was much higher than that in HIF-1α negative group (13.7±3.9) (P 〈 0.05). MVD was positively correlated with the expression of HIF-1α (r =0.901 ,P 〈 0.05). HIF-1α expression was not correlated with the extent of tumor differentiation, age, gender and tumor region. Conclusion HIF-1α expression may play an important role in the infiltration and metastasis of the laryngeal spquamous cell carcinoma.
