28味中药酸性成分提取物影响黑素合成的实验研究 被引量:22

Studies on effect of acidic extracts from 28 kinds of Chinese materia medica on melanin synthesis

摘要 目的:证实中药酸性成分对黑素的产生具有抑制作用。方法:采用蘑菇酪氨酸酶多巴速率氧化法,体外测定28味中药酸性成分高、中、低浓度对酪氨酸酶活性的影响。结果:①28味中药酸性成分在高浓度(25g/L)时对酪氨酸酶活性均具有抑制作用,其中乌梅抑制率最高,达96.50%;五味子次之为85.33%,与2mol/L的熊果苷抑制作用(56.67%)相比有极显著差异(P<0.01);地肤子(81.66%)、枇杷叶(81.65%)等8味中药酸性成分有显著差异(P<0.05)。②19味中药酸性成分高、中、低浓度对酪氨酸酶活性呈剂量依赖性抑制作用。③甘草、枸杞子、五味子等8味中药酸性成分对酶活性呈高浓度抑制,中、低浓度激活的双向作用。结论:中药酸性成分提取物可抑制黑素的合成,其机制与抑制酪氨酸酶活性有关。 Objective: To confirm acidic extracts of Chinese materia medica having an inhibition to melanin synthesis. Methods: By L-DOPA oxidation method, the in vitro determination was studied to get tyrosinase activity's inhibition of acidic extracts from 28 kinds of Chinese materia medica with high, middle and low concentration solution. Results: First, Acidic extracts from 28 kinds of Chinese materia medica at high concentration (25g/L) showed potent inhibitory action on tyrosinase activity. The highest inhibition rate was 96.50%, which was from the acidic extract of Fructus Mume. The next was fructus schizandrae with 85.33% inhibition rate. Compared with a 56.67% inhibition rate of 2 mmol/ml control sample arbutin, both Fructus Mume and fructus schizandrae had a large significant deviation ( P〈 0.01). Other 8 acidic extracts from Fructus Kochiae (81.66%), Folium Eriobotryae (81.65%) and so on also had a significant deviation ( P〈 0.05). Second, 19 kinds of acidic extracts at high, middle and low concentration showed potent inhibitory action on tyrosinase activity and the effectiveness of inhibition was proportional to their concentration. Third, 8 kinds of acidic extracts, such as Radixl Glycyrrhizae, Fructus Lycii and fructus schizandrae etc, had a potent inhibition to tyrosinase activity at high concentration and showed a two-way activation at middle and low concentration. Conclusion: Acidic extracts of Chinese materia medica can inhibit melanin synthesis, which mechanism was related to the inhibition of tyrosinase activity.
