摘要
目的:探讨腺嘌呤致大鼠少精子症模型的最佳剂量和造模时间点。方法:用0.5%羧甲基纤维素钠溶液,将腺嘌呤按0、20、25、30、35mg/ml浓度配制,75只大鼠按体重随机分为5组(0、200、250、300、350mg/kg),每组15只,按1ml/100g灌胃,1次/日。造模10d、20d、30d各处死一批,共3批。观察大鼠一般状态,取附睾尾作精子计数及形态学观察,取血测肝、肾功能及生殖激素,称量肝、肾及生殖器官湿重并计算脏器指数,切片进行病理形态学观察。结果:各模型组相继出现不同程度的肾阳虚体征;尿素氮(BUN)、肌酐(CRE)第10d开始明显升高;20d开始血清睾酮明显下降;随着造模时间的延长和剂量的增加,精子密度和活力出现不同程度的降低,畸形率上升,并呈一定的时效和量效关系。肝、肾指数明显升高,生殖器官指数明显降低;造模30d时,多数模型组睾丸内生精细胞和附睾精子数量减少。结论:腺嘌呤致大鼠少精子症模型的最佳剂量为250mg/kg,造模时间点为20d。
Objective : To investigate optimal dose and duration for rat model with oligozoospermia induced by adenine. Methods:Adenine was dissolved with 0.5% solution of sodium carboxymethyl cellulose (CMC) and was prepared at the concentration of 0, 20, 25, 30 and 35mg/ml. Seventy-five rats were randomly assigned to 5 groups ( 15 rats in each group) according to body weight of rats and received intragastric adminis- tration of the adenine solution at lml/100g body weight of rats per day. Five rats from each group were sacri- ficed on the 10th, 20th and 30th day after the first administration of adenine. The general living condition of rats was observed. Epididymal sperm concentration and morphology, hepatic and kidney functions, and re- productive hormones were evaluated. The wet weight of liver, kidney and reproductive organ of rats were assayed, using for calculating the viscera index. In addition, pathological changes were examined. Results: Signs of kidney - yang deficiency at different degree gradually appeared after the administration of adenine. The serum creatinine (CRE) and blood urea nitrogen (BUN) began to notably increase on the 10th day after the first administration of adenine. The serum testosterone (T) concentration and the reproductive organ/ body weight ratio were significantly decreased while the liver and kidney index were obviously increased on the 20th day after the first administration of adenine. With the increase of the duration and dose of adenine administration, epididymal sperm concentration and motility decreased while sperm abnormality rate in- creased. The amount of sperulatogenetic ceils in the testes and sperm in the epididymis reduced on the 30th day after the first administration of adenine. Conclusion : The optimal dose and duration for rat model with oli- gozoospermia induced by adenine are 250 mg/kg and 20 days respectively.
