妊娠期间脾脏T淋巴细胞的改变与鼠胚着床

摘要 应用FITC标记的L3T4及Ly2单克隆抗体,及流式细胞计细胞分选,对小鼠妊娠期间脾脏T淋巴细胞亚群进行检测。结果表明,妊娠早期小鼠脾脏CD8+淋巴细胞比例减少,在着床时尤为明显。提示脾脏T淋巴细胞数量和功能变化,参与维持正常妊娠,尤其是参与着床。 The functional role of the spleen in pregnancy is not clear. They may regulate the maternal immune response to the fetal allograft. This study was designed to study the change of T lymphocyte subpopulations in pregnant mouse spleen and compare them with that fo non pregnant mouse.Methods:Spleens were obtained from pregnant D2,D6,D12,D18 and postpartum pregnant mice as well as non pregnant mice as control. Lymphocytes were prepared to the required concentration of 1×106 cells/ml and labelled withanti L3T4 FITC or anti Ly2 FITC directly.Samples were analyzed by a fluorescence activated cell sorter (Becton Dickinson FACS can).Results:the percentage of CD*+lymphocyte decreased during normal pregnancy as compared with normal non pregnant mice. The decrease in the CD8+lymphocyte jusy occured from implantation (D6) and showed most significantly(13.88±1.9135 versus 21.13±0.5726,P<0.001).Thisdecrease remained in the second trimester (D12)(15.68±0.9155 versus 21.13±0.5726,P<0.001) and the third trimester (D18)(16.31±1.4994 versus 21.13±0.5726,P<0.001).In postpartum mice, the decreased percentage of CD8+lymphocyte rise, to the level of near control. No significant relation was found between CD4 lymphocyte subpopulations of pregnancy and that of control group. CD4+/CD8+ratio increased from inplantation accordingly. All these findings suggest that the changes of CD8+lymphocyte subpopulations may play an important role in maintaining normal pregnancy and especially, implantation.
