p38MAPK在慢性髓细胞白血病信号转导中的作用

摘要 目的:探讨p38MAPK在慢性髓细胞白血病(CML)信号转导中的作用。方法:用脂质体转染法将bcr3/abl2反义寡核苷酸(ASO)导入K562细胞中,通过细胞凋亡、p38MAPK活性、p38MAPK表达和p38MAPK含量的变化,检测p38MAPK在CML信号转导中的作用。结果:bcr3/abl2ASO作用K562细胞24h后凋亡细胞比率、p38MAPK活性、p38MAPK表达和p38MAPK含量明显增加,分别为对照组的3.6、3.3、2.7和1.8倍,与对照组之间均差异有统计学意义(均P<0.05)。结论:p38MAPK通过介导凋亡信号而在CML信号转导中起重要作用。 Objective:To study the role of p38 mitogen-activated protein kinase(p38MAPK) in the signal transduction of chronic myelogenous leukemia(CML).Method:After Bcr3/abl2 antisense oligodeoxynucleotide(ASO) was introduced into K562 cell line by liposomal transfection,the effect was evaluated by apoptosis,p38MAPK activity,p38MAPK expression and p38MAPK content.Result:After 24 hours treatment by ASO,the rate of apoptotic cells,p38MAPK activity,p38MAPK expression and p38MAPK content were 3.6,3.3,2.7 and 1.8 times higher than that in control groups respectively(P〈0.05).Conclusion:p38MAPK plays an important role in the signal transduction of CML by directing apoptotic signal.
