摘要
目的:研究母体暴露壬基酚对仔鼠学习记忆及脑组织AchE和ChAT活性的影响。方法:母鼠孕9~15d灌胃壬基酚(0、50、100、200mg/kg),用Morris水迷宫法观察壬基酚对仔鼠学习记忆的影响;检测F1仔鼠垂体和海马组织乙酰胆碱脂酶(AchE)和胆碱乙酰转移酶(ChAT)的活性。结果:与阴性对照组比较,NP200mg/kg组仔鼠水迷宫中逃避潜伏期延长,脑组织ChAT活性明显下降、AchE活性上升(P<0.05)。结论:壬基酚可透过胎盘屏障影响仔鼠学习记忆功能,机理可能与影响胆碱能神经系统功能有关。
Objective:To investigate the effects on the activity of acetylcholinesterase (AchE) and choline acetylase(ChAT) in filial rat’s brian and learning and memory capacity after the mother-rat is exposed to nonylphenol. Methods:At the first day of the pregnancy,the rats were divided into four groups,which were orally administered with NP at doses of 0,50,100 and 200 mg/kg,respectively,on gestational days 9~15. The offspring rats’s Brain tissues of each groups were dissected for AchE and ChAT activity assay and cognitive function was tested by Morris water maze. Result:Morris water maze test results showed the escape latency was longer in the groups of NP 200 mg/kg than in control group(P〈0.05).The AchE activity decreased while the activity of ChAT increased in the NP 200 mg/kg group, the differences mentioned above were statistical significant compared to control group,with dose-effect relationship in some degree, (P〈0.05).Conclusion:Exposures to NP during gestation might influenced the activityof AchE and ChAT in brian of F1 rats .
