甲状腺过氧化物酶显示电镜方法的改进

摘要 目的:探讨甲状腺过氧化物酶电镜显示方法的改进。方法:介绍4项改进方法,即液氮骤冷、漂浮固定、漂浮孵育、倒扣包埋。结果:改进后的方法在透射电子显微镜下能清晰、准确地观察到经DAB方法显示出甲状腺组织的滤泡上皮细胞顶端膜,高尔基复合体及微绒毛过氧化物酶阳性反应。 Objective:The method improvement for the thyroid peroxidas shows under the electron microscope in four parts:(1)liquid instant cold, (2)flotation fixation method(3)flotation method incubation (4)stand upside down embedding.Results:Under the electron microscope, the improved method can provide a clear and accurate observation of the follicle epithelial cell membrane of thyroid tissue displayed by DAB methods, and shown the positive reaction of Golgi complex and microvill. us the thyroid peroxidase as well.Conclusion:This technique used as a sign by there enzyme fixed displayment provides a reliable and base theory for the study of physiological and pathological change of the thyropathy.
