摘要
目的探讨椒目油对支气管哮喘(哮喘)豚鼠模型肺泡灌洗液(BALF)中IL-4、IFN-γ水平及肺组织中NF-κB的影响,为椒目油治疗哮喘提供理论依据。方法实验分4组:正常对照组、哮喘模型组、地塞米松治疗组和椒目油治疗组,每组各10只豚鼠。用卵白蛋白(OVA)腹腔注射致敏和雾化吸入激发建立哮喘豚鼠模型。ELISA法检测豚鼠BALF中IL-4,IFN-γ含量。免疫组化法观察NF-κB在豚鼠支气管上皮的表达。结果哮喘组BALF中的IL-4、IFN-γ含量及PC_(20)水平都与正常对照组有显著的差别(P均<0.05),分别为(41.36±6.71)ng/L和(10.58±1.28)ng/L,(21.15±2.75)ng/L和(73.52±5.23)ng/L,(0.013±0.014)g/L和(0.168±0.186)g/L,而椒目油组及地塞米松治疗组豚鼠BALF中IL-4含量分别为(15.35±4.28)ng/L、(19.20±3.78)ng/L,明显低于哮喘模型组(P<0.05),其IFN-γ含量分别为(50.65±4.19)ng/L、(53.34±5.64)ng/L和PC_(20)[(0.160±0.180)g/L、(0.144±0.154)g/L]水平较后者显著升高(P<0.05)。椒目油治疗组和地塞米松治疗组各指标之间无显著性差异(P>0.05)。正常对照组、哮喘模型组、椒目油治疗组和地塞米松治疗组豚鼠气道中NF-κB阳性细胞百分比分别为(9.51±2.82)%、(52.71±7.80)%、(32.26±3.70)%、(29.89±2.01)%,两治疗组均和哮喘模型组有显著差异。结论椒目油能有效降低豚鼠BALF中IL-4水平,升高IFN-γ含量,同时降低气道高反应性和NF-κB在哮喘豚鼠支气管上皮中的表达。
Objective To explore effects of Jiaomu Oil on IL- 4 and IFN-γ in bronchoalveolar laVage fluid(BALF) and NF-κB in the lung tissue of asthmatic model in guinea pig and to provide theoretical basis for treatment of bromchial asthma (asthma) using Jiaomu Oil. Methods This experiment included four groups:normal control group, asthmatic model group, dexamethasone treatment group and Jiaomu Oil treatment group, involving 10 guinea pigs in each group. Asthmatic models of guinea pigs were constructed by immunization of intraperitoneal injection with saline ovalbumin(OVA) and challenge of inspiration with nebulized OVA. The levels of IL-4 and IFN-γ in BALF of guinea pigs were determined using enzyme-linked immunosorbent assay ( ELISA ). The expression of NF-κB in the lung tissue was observed by immunohistoehemical staining. Results In the asthmatic model group,the levels of IL-4 and IFN-γ in BALF and PC20,(41.36±6.71) ng/L vs (10.58±1.28) ng/L,(21.15±2.75) ng/L vs (73.52±5.23) ng/L, (0. 013±0. 014) g/L vs (0. 168±0. 186) g/L, had significant diiferences compared with those of normal control group, respectively (P 〈 0.05). The levels of IL 4 in BALF of Jiaomu Oil and dexamethasone groups,(15.35±4.28) ng/L and (19.20 ± 3.78) ng/L, were both significantly lower than those of asthmatic model group( P 〈20.05). But the levels of IFN-γ in the above two groups,BALF, (50.65±4.19) ng/L,(53.34±5.64) ng/L and PC20 (0.160±0.180) g/L,(0.144±0. 154) g/L,were significantly higher than those of asthmatic model group ( P 〈 0.05). The levels of IL- 4, IFN y and PC20 did not have any statistical significance between Jiaomu Oil and dexamethasone groups ( P 〉 0.05). The percents of NF-κB positive cells in normal group, asthmatic model group, Jiaomu Oil group and dexamethasone group were (9.51±2.82)% ,(52. 71±7. 80)% ,(32.26±(3.70) % and (29. 89±2.01)% respectively. The percents of two treatment groups both had statistical significance compared with that of asthmatic model group. Conclusions Jiaomu Oil can efficiently decrease the levels of IL -4 and increase IFN -γ in BALF, it can decrease airway hyperresponsiveress and expression of NF-κB in the asthmatic guinea pig.
