布鲁氏菌蛋白变应原的实验及应用研究

目的：解决人用布鲁氏菌病皮内诊断制剂的非特异反应及毒副作用问题。方法：用三氯醋酸和硫酸铵从布氏菌菌液中制备了纯化的布氏菌蛋白变应原，对动物及布病病人、布氏菌隐性感染者、布氏菌苗免疫者、肺结核和其他病人及健康人群做皮内变态反应试验。结果：该制品具有良好的皮肤变态反应原性，其特异性和敏感性与国际相关标准品完全一致；现场研究结果显示，其显著特点是安全性好，无副作用，非特异性反应大大低于现行国产市售菌素（速发反应少，反应面积小）；在保证该变应原具有良好的特异性和敏感性的前提下，提出了阳性判定标准。结论：该蛋白变应原为纯度高、质量好、易标化的新型布鲁氏菌病诊断制剂。

Objective:In order to solve the nonspecific reaction and poisonous side effect problem from brucellosis cutireaction diagnostic preparation for human beings.Methods: We have prepared puried Brucella protein allergen from Brucella cultures with trichloracetic acid and ammonium sulphate. This kind of allergen has been utilized in cutireation test for animals, brucellosis patients, brucellar recessive infective crowd, immunized crowd with brucellar vaccine, pulmonary tuberculosis, other kinds of patients and healthy crowd. the experiment results prove that this kind of allergen has good original metamorphotic reaction, the specificity and sensibility is consistent with the interrelated standardized product in the world. Results: The testing ground results proved that its notable characteristic was good security and no side effect, and its nonspecific reaction was far less than the current national brucellin(showed in bless rapid metamorphosis reaction and smaller reaction area). So we put forword a positive judgment standard under the prerequisite for the allergen keeping good specifity and sensibility. Conclusion: This kind of protein allergen is a newtype high-purity, good-equality and easy-standardization diagnostic preparation for brucellosis.
