摘要
目的观察密蒙花提取物滴眼剂对去势所致干眼症雄鼠基础泪液分泌量、泪膜稳定性以及泪腺电镜显微结构的影响。方法将45只(90眼)Wister雄性大鼠随机分为空白组(A1、A2、A3)、模型组(B1、B2、B3)、密蒙花组(C1、C2、C3)共9小组,1、2、3分别代表给药时间为1月、2月、3月,每组5只(10眼),对B、C组行去势术建立动物模型,对C组以密蒙花提取物滴眼剂连续滴眼1月,对全部实验大鼠行Schirmer I试验、测量泪膜破裂时间,电镜观察泪腺超微结构变化。结果C组Schirmer I试验测量值明显高于B组,差异具有统计学意义(P﹤0.01),泪膜破裂时间明显长于B组,差异具有统计学意义(P﹤0.01),显示密蒙花提取物滴眼剂能够显著维持泪腺基础分泌量和泪膜的稳定性;电镜示C组显微结构明显改善。结论密蒙花提取物滴眼剂中主要成分为黄酮类物质,可显著改善泪腺组织超微结构,维持泪腺基础分泌量和泪膜的稳定性。
Objective To assess the effects of eye drops with the extract of buddleja officinalis on secreting volume of basic tears, stability of tear film and the microstructure of lacrimal gland under electronic microscopy in castrated rats with dry eye. Methods 45 Wister rats were divided averagely into 9 groups,including normal group(A1 ,A2 and A3), model group (B1,B2 and B3),Mimenghua group (C1,C2 and C3), 5 rats with 10 eyes per group. The"1", "2" and "3" respectively meant treated times in 1 month, 2 months and 3 months. The dry eye model was established with orchiectomy in Group B and C. the rats in group C were treated with buddleja officinalis extract eye drops. For all rats, SIT and BUT were made, morphological and ultrastructure changes were observed by electron microscopy. Results The SIT value in group C was significantly higher than that in group B (P〈0.01) and the BUT value in group C was significantly longer than that in group B(P〈0.01), which showed that the eye drops could significantly keep basic tears secretory volume and tear film stability, and the ultrastructure in C group was improved. Conclusion The main components of buddleja officinalis extract is the flavonoids, which can improve ultrastructure of lacrimal gland and keep the stability of basic tears secretory volume and tear film.
