摘要
本试验收集了临床发生黄疸病犬的尿液,提取基因组DNA,经16s rRNA基因PCR鉴定,表明该犬存在钩体黄疸出血群毒株感染。以该基因组DNA为模板,扩增获得lipL32基因,并对该基因进行了原核表达,结果显示该抗原在大肠杆菌中获得高效可溶性表达。该蛋白的成功制备为犬钩体的诊断及新型亚单位疫苗的研制提供了广谱候选抗原。
In the present study, urine samples were collected from a dog infected with Leptospira interrogans serogroup Icterohaemorrhagiae and used as the genomic template for DNA extraction. The 16 s rRNA and lipL32 gene was amplified and subcloned into pET32a ( + ) vector for expression in E. coli BL21 (DE3) strain. Results showed that lipL32 gene was highly expressed and the product was soluble. The availability of LipI.32 protein will benefit for development of diagnostic reagents and novel subunit vaccine against canine ieptospirosis.
